چکیده

To investigate the anti-fibrotic effects of Astragaloside IV in rats with experimental colitis, and to explore the possible mechanisms, Sprague-Dawley Rats with colitis were induced by 2,4,6-trinitrobenzene sulphonic acid (TNBS). Forty-eight rats were randomly divided into normal group, TNBS model group, SB203580 group and Astragaloside IV group. 150 mg/kg TNBS dissolved in 50% ethanol were gavaged into each rat in the three groups but not the normal group. Rat in the normal group was gavaged with an equal volume of 0.9% NaCl solution. Rats in the SB203580 group were intraperitoneally injected with 10 mg/kg of SB203580 daily for 4 days. Rats in the Astragaloside IV group were gavaged with 25, 50 or 100 mg/kg of Astragaloside IV daily for 21 days. At the end of the experiment, colon tissue samples were collected, and the injury and fibrosis of the colon were detected by HE staining and Masson collagen staining, respectively. Expression of transforming growth factor (TGF)-β1, connective tissue growth factor (CTGF), collagen I, collagen III, fibronectin (FN) and α-SMA was determined by western blotting. Compared with the TNBS model group, the histological scores and fibrosis were improved significantly in the SB203580 group as well as Astragaloside IV group. Expression of TGF-β1, CTGF, Collagen I, Collagen III, FN and α-SMA in colonic tissues in the SB203580 group and Astragaloside IV group was significantly decreased compared with that in the TNBS model group. In conclusion, Astragaloside IV reduces intestinal fibrosis in rats with TNBS-induced colitis.

Astragaloside IV ameliorates 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced colitis implicating regulation of energy metabolism

Dysfunction of energy metabolism is involved in inflammatory bowel disease (IBD). This study was designed to investigate the potential of astragaloside IV (ASIV), an active ingredient of Radix Astragalus, to ameliorate colonic mucosal injury, with focusing on the implication of energy restoration in the underlying mechanism. Experimental colitis model was established in rats by injecting 2,4,6-...

Objective(s):In the present study, we evaluated immunological and immunomodulatory properties of royal jelly (RJ) in 2,4,6 trinitrobenzene sulfonic acid (TNBS)-induced colitis in rats. Materials and Methods: Eighteen adult female Wistar albino rats were divided into three groups of six animals each: a control group that received only saline solution, a TNBS-induced colitis group, and a TNBS-co...

Glutamine prevents fibrosis development in rats with colitis induced by 2,4,6-trinitrobenzene sulfonic acid.

We investigated the effects of glutamine on the development of colonic fibrosis and on the expression of the major fibrogenic factors in a rat model of experimental colitis. Colitis was induced in one-half of the male Wistar rats by intracolonic administration of 30 mg of 2,4,6-trinitrobenzene sulfonic acid (TNBS). L-glutamine (25 mg/kg) was administered rectally to one-half of the controls and...

Objective(s): This study was aimed at investigating immune activations of the 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced colitis model in colonic mucosa by immunohistochemical and Western blot methods. Materials and Methods: For this purpose, 16 female Wistar albino rats were divided into two random groups of control (n=8) and colitis (n=8). The experimental colitis model was induced by...

Magnetic Resonance Colonography for Fibrosis Assessment in Rats with Chronic Colitis

BACKGROUND Magnetic resonance colonography (MRC) has been developed to assess inflammatory bowel diseases. We aimed to assess the feasibility of MRC in rats with TNBS-induced chronic colitis and to confront imaging results with fibrosis and stenosing features of the model. MATERIALS AND METHODS Chronic colitis was induced in 12 rats by weekly intra-rectal injection of increasing doses of TNBS...
